pPsiR from Sinorhizobium fredii
This part is the promoter region (0.4 kb upstream) of the PsiR gene of Sinorhizobium fredii (NGR_b11520)
pPsiR is predicted to be repressed by the PsiR transcription factor (BBa_K2448007) which is inhibited in the presence of D-Psicose [1].
Usage and Biology
Our team used this pPsiR promoter together with the Sinorhizobium fredii PsiR transcription factor (BBa_K2448007) to build a Psicose biosensor (BBa_K2448029). The results show that pPsiR is functional in E. coli and that it behaved as predicted under and without induction.
Using the BPROM webserver [2], the predicted features of this promoter are: Threshold for promoters - 0.20 Number of predicted promoters - 1 Promoter Pos: 350 LDF- 1.71 -10 box at pos. 335 gactatcct Score 43 -35 box at pos. 314 ttgcac Score 33 Oligonucleotides from known TF binding sites: No such sites for promoter at 350
